p53 is a tumour suppressor protein which is the most commonly mutated protein in human tumours. It is therefore of considerable interest to an understanding of why some cells develop into cancer cells leading to the formation of tumours. These mutations make this protein dysfunctional in over 50% of human tumour samples from which it has been sequenced. A functionally important part of the structure of p53 is the tetramerization domain which includes residues 325 355. In intact p53, this domain is responsible for forming a tetramer between four individual polypeptides. Even when expressed as a short peptide, this tetramerization activity is conserved resulting in the smallest known protein tetramer. In this example, CD was used together with uorescence (Section 3.4) and differential scanning calorimetry (DSC; 8) in a study of the structural stability of this tetramerization domain. The following plot shows how ellipticity of a small concentration (2.5 M) of the protein varies with temperature. The intersection of all the traces at a single point called the isodichroic point is characteristic for a two-state system such as a protein only existing in either a folded or unfolded state (Section 3.4.5). As the temperature is increased from 21 to 81 C, the ellipticity measured in the region of 220 nm decreases closer to 0. Such measurements allow calculation of the fraction of the protein unfolded. This allowed determination of unfolding curves at various concentrations of protein in the range 0.5 20 mM as shown in the following plot.
